HybBox™ is intended to use for hybridization
of denatured sample slides during FISH (or ISH) process. HybBox™ is an insulated
EPS (expanded polystyrene) box that consists of two pieces: lid and base. After
denaturation of sample slides on metal slide tray in oven, the hot metal tray
with slides is transferred into the base of HybBox™. Then, the box is closed tightly
with its lid. Well insulated HybBox™ preserves effective hybridization. The HybBox™
should not be opened until post-hybridization washing is ready.
Hybridization time which is required to produce desired signals with repetitive
satellite DNA probes may vary from 30 min (cell FISH) to 2-3 hours (tissue FISH).
For FISH on cells or paraffin-embedded tissue sections with chromosome paint and
unique sequence probes, hybridization for 3-4 hours in HybBox™ is sufficient to
obtain desired signals, but overnight hybridization is recommended for paraffin-embedded
tissue sections in order to produce stronger signals.
Hybridization in HybBox™ is more convenient than in conventional incubator. Hybridization
may be performed conveniently overnight in HybBox™ until post-hybridization washing
is ready. Extended or overnight hybridization in HybBox™ generally provides brighter
signals. HybBox™ is light and portable. Multiple HybBox™ boxes can be stacked up
for processing numerous sample slides.
